CONTRIBUTION OF THE PENTOSE CYCLE TO THE METABOLISM OF GLUCOSE IN THE ISOLATED, PERFUSED BRAIN OF THE MONKEY

Abstract— Isolated brains from three adult monkeys were perfused for 1 hr with [2-¹⁴C]glucose. Glycogen was isolated from the brain stem, cerebral hemispheres, cerebellum and the hypothalamic area at completion of the perfusions. The distribution of ¹⁴C in carbons of the glucose unit of glycogen was determined and from this the contribution of the pentose cycle to metabolism of glucose was calculated. The data indicate a maximum contribution by the pentose cycle of 5–8 per cent in brain. No significant difference was observed in the various portions of brain. Oxygen consumption was noted to be low in relation to the amount of glucose utilized, as measured in these experiments.     

Improved isolation of a 50 kDa anorexigenic protein from rat urine

In prior work, a 50 kDa protein was purified to homogeneity from rat urine. This protein reduces food intake when injected into rats and is the only natural substance other than satietin known to be effective for long (24 hour) time periods and which does not make animals ill. However, when attempts were made to repeat the purification, contamination appeared in the 50 kDa fraction. The present contribution documents successful reisolation of the 50 kDa anorexigen by an improved method. Reisolation involved Cibacron blue-Sepharose, DEAE-Sephacel and Sephacryl S-200 chromatography, and SDS disc preparatory electrophoresis. The reisolated 50 kDa anorexigen contains no detectable carbohydrate. Partially purified preparations of the 50 kDa anorexigen were fragmented with trypsin and proteinase K without loss of anorexigenic activity. It is concluded that the 50 kDa anorexigen may be reproducibly purified to homogeneity and may contain within its amino acid sequence a peptide which is the basis of its anorexigenic activity.     

Role of virus inhibitory factor or interferon in the antitumoral effect of whole-body hyperthermia

The interferon inducing effect of hyperthermia was studied in normal and tumor-bearing mice. Circulating interferon was temporarily detected one day after subcutaneous transplantation of 1.6 X 10(6) Ehrlich's ascites tumor cells. Hyperthermia of 43.5 degrees C for 5 min did not induce the interferon formation in mice with or without subcutaneous tumor of the cells. These findings showed that the induction of interferon formation was not main cause of the hyperthermia-induced tumor inhibition.     

Androgen-dependent messenger RNA(s) related to secretory proteins in the mouse epididymis

Total RNA from mouse epididymides was translated in a cell-free system derived from rabbit reticulocyte lysate. The androgen dependence of a highly represented mRNA(s) was detected. This mRNA(s) encoded for a band of Mr 26,000 visualized by denaturing gel electrophoresis (SDS-PAGE). No other mouse sexual tissues (testis, vas deferens and seminal vesicle), liver, kidney or striated muscle presented such a band. Furthermore, this mRNA(s) was restricted to the caput epididymidis. Two-dimensional gel electrophoresis (2D-PAGE) showed that this band of Mr 26,000 was composed of 6 basic polypeptides. This translated protein may correspond to a newly synthesized secretory protein of Mr 24,000. The concentration of translatable mRNA(s) encoding for the band of Mr 26,000 dropped to 31% of the normal level at 3 days after castration and to 7%, 20 days later. Administration of testosterone to 30-day-castrated mice partly reversed these changes: 32% of the control value was reached after 10 days of treatment. Accumulation of this mRNA(s) was also just detectable (7% of the mature level) at 10 days of age. The high levels attained by 20 days of age (60% of the control value) suggest that the greatest increase of accumulation for this mRNA(s) occurs during the prepubertal period.     

Reduced sensitivity of peripheral cells to glucocorticoids in hypercholesterolemia

It has been found that lymphocytes of hypercholesterolemic (HCh) subjects are characterized by a reduced number of glucocorticoid receptors (GcR) as compared with the cells of normolipidemics (N). Addition of HCh-sera or very low density lipoproteins, or low density lipoproteins isolated both from HCh-sera and N-sera to cultured human skin fibroblasts brought about a fall in the number of GcR in the cells. High density lipoproteins had no effect on GcR level. Dexamethasone was less effective in inhibiting cholesterol synthesis from [14C]acetate in the lymphocytes and fibroblasts with a reduced number of GcR. In the presence of dexamethasone (I x 10(-8)M) in fibroblast growth medium, reduced number of GcR (due to preincubation with very low density lipoproteins) led to a substantial increase in cholesterol synthesis. These findings indicate that the sensitivity of peripheral cells to glucocorticoids is decreased in HCh which might be one of the trigger mechanisms of atherogenesis.     

Self-complementarity of messenger RNA's of periodic proteins

Summary It is shown that the mRNA's of three periodic proteins, collagen, keratin and freezing point depressing glycoproteins show a marked degree of self-complementarity. The possible origin of this self-complementarity is discussed.
